摘要

目的:探索甘草查尔酮A对人口腔鳞癌细胞自噬活动的影响.方法:分别采用0、25、50 μmol/L 甘草查尔酮A刺激人口腔鳞癌细胞SCC-25系细胞,并分别于刺激后0、6、24 h收集细胞,采用流式细胞计数检测溶酶体数量,采用Real-time PCR、Western blot及免疫荧光染色检测自噬相关分子基因及其蛋白的表达情况.结果:甘草查尔酮 A可呈剂量及时间依赖性的促进SCC-25鳞癌细胞自噬活性,表现为促进其溶酶体及自噬泡的形成、促进其表达自噬调节因子beclin1及LC3,且显著增高LC3-Ⅱ/LC3-Ⅰ的比率(P<0.05).未经刺激组SCC-25系细胞基本不表达LC3蛋白,而经50 μmol/L甘草查尔酮A刺激后,LC3蛋白表达显著增强(P<0.05),且主要表达在胞浆.结论:甘草查尔酮 A可呈时间、剂量依赖性的促进人口腔鳞癌细胞的自噬活动,这可能是甘草查尔酮 A抗肿瘤作用的重要机制.

Abstract

AIM:To investigate the effects of licochalcone A on the autophagy of human oral squamous cell carcinoma cells (OSCCs).METHODS:OSCC SCC-25 cells were treated once with 0,25 and 50 μmol/L of licochalcone A for 0,6 and 24 h respectively,and then the cells were harvested for further autophagy detection.The amounts of lysosomes were quantified by flow cytometry,and the gene and protein level of autophagy-related molecules were evaluated by real-time PCR,western blot and immunohistochemistry.RESULTS:Licochalcone A dose-and time dependently increased the autophagic activities of SCC-25 cells,characterized by the formation of lysosomes and autophagic vacuoles,increased gene and protein expression of beclin1 and LC3,and increased LC3-Ⅱ/LC3-Ⅰ ratio (P<0.05).No LC3 protain expression was observed in untreated SCC-25 cells,while after 50 μmol/L licochalcone A treatment high level of LC3 protein in SCC-25 cells was observed,predominantly in cytoplasm (P<0.05).CONCLUSION:Licochalcone A may promote the sautophagic activitiy of SCC-25 cells,which might be one of the important mechanisms of its anti-tumor effects.
