摘要

目的:利用同轴静电纺丝技术制备骨保护素-聚乳酸(OPG-PLA)/壳聚糖电纺纤维膜,评价其相关理化性能.方法:分别制备同轴OPG-PLA/壳聚糖电纺纤维膜和PLA/壳聚糖电纺纤维膜.SEM观察两种纤维膜表面形貌并测量纤维直径,透射电镜鉴定同轴结构,生物力学测试仪测量纤维膜力学性能,并测定纤维膜降解质粒释放率.结果:所制备的同轴OPG-PLA/壳聚糖电纺纤维与PLA/壳聚糖电纺纤维直径均匀,纤维光滑,方向随机,无串珠.同轴OPG-PLA/壳聚糖电纺纤维和PLA/壳聚糖电纺纤维的直径分别为(468±53)nm和(375±65)nm(P<0.05),拉伸强度分别为(1.85±0.37)MPa和(4.23±0.51)MPa(P<0.05)、弹性模量分别为(12.58±1.73)MPa和(13.26±2.79)MPa(P>0.05),质粒释放较平稳,可持续30 d 以上.结论:采用同轴静电纺丝法制备OPG-PLA/壳聚糖电纺纤维膜形态良好,力学性能符合要求,具有缓释效果.

Abstract

AIM:To prepare a novel osteoprotegerin(OPG)-polylactide (PLA)/chitosan(CS)nano-fibrous membrane by coaxial electrospinning,and to evaluatethe biological properties of the OPG-PLA /CS fiber.METHODS:A nanofibrous membranes with OPG-PLA/CS and PLA/CS were respectively prepared by coaxial electrospinning method.The morphology,mechanical properties and plasmid release rate of the membranes were observed and evaluated.RESULTS:The OPG-PLA/CS fiber diameter was uniform and the surface of the fiber was smooth.The diameter of OPG-PLA/CS and PLA/CS was(468±53)nm and (375±65)nm(P<0.05),the tensile strength was(1.85±0.37)MPa and(4.23±0.51)MPa(P<0.05),the Young's modulus was (12.58±1.73) MPa and (13.26±2.79)MPa (P>0.05),respectively.The release of plasmid was going on for more than 30 d and stable.CONCLUSION:The OPG-PLA/CS fiber prepared by coaxial electrospinning method may have excellent mechanical property and controlled release of plasmid.
