摘要

目的:探讨建立正畸小鼠牙齿移动模型的有效方法.方法:选用雄性10周龄C57小鼠30只,随机分为0、1、3、5、7 d组(n=6).除0 d组外每只小鼠上颌右侧切牙与第一磨牙之间安置正畸拉簧,并在加力过程中定期观察其脱落情况,计算脱落率.分别在相应时间处死各组小鼠,取实验侧标本,CT观察牙齿移动状况.标本脱钙后冰冻切片,HE和免疫荧光染色观察骨质改建情况.结果:CT结果显示,第一磨牙和第二磨牙之间从第1天开始出现间隙,随时间延长间隙逐渐加大,HE染色显示压力侧存在骨质改建;免疫荧光trap染色显示压力侧存在破骨细胞且膜腔变窄.结论:小鼠牙齿移动模型制作成功.

Abstract

AIM: To establish mouse model of orthodontic tooth movement.METHODS: 30 male C57 mice aged 10 weeks were randomly divided into 0,1,3,5 and 7 d groups(n=6).The right side of jaws attached with orthodontic appliances were used as the experiment group.In the movement progress,the mice were regularly checked and the loss rate of the orthodontic tension spring was calculated.The mice in each group were executed at the corresponding time,the iaw samples were taken and fixed,the movement of the teeth was observed by CT scanning.HE staining and immunofluorescence staining were used to observe the bone remodeling.RESULTS: CT results showed that the teeth were obviously moved.HE staining showed that there was bone remodeling in the pressure side,and immunofluorescence staining showed that at the pressure side there the periodontal membrane became narrow,osteoclasts were observed.CONCLUSION: The mouse model of orthodontic tooth movement is established.
