Objective To apply fluorescence polarization method to detect human serum brucellosis(referred to as"brucellosis")antibody,analyze its application value in the diagnosis of brucellosis,so as provide scientific basis for laboratory detection of brucellosis.Methods A total of 245 sera from key surveillance of population of brucellosis in Guizhou Province in 2021 were collected and simultaneously used fluorescence polarization method(FPA),rose bengal test(RBT),serum agglutination test(SAT)and enzyme-linked immunosorbent assay(ELISA)for detection.The consistency,sensitivity and specificity between the results of the four methods were analyzed.Results The Kappa values of FPA compared with RBT,SAT and ELISA were 0.852,0.845 and 0.778,respectively.The Kappa values of FPA and traditional RBT and SAT were all more than 0.750.This means that there was a high consistency between FPA and the other three diagnostic methods.The Kappa values of ELISA and RBT,SAT and FPA were 0.779,0.804 and 0.778,respectively,and the Kappa values for ELISA versus and the other three methods were between 0.610 and 0.800,ELISA was consistent with the other three diagnostic methods.The sensitivity and specificity of RBT and confirmed to confirmatory test SAT were higher.The sensitivity and specificity of FPA and ELISA were also higher than that of SAT,with a sensitivity of 86.2%,but the specificity of FPA was 98.4%,higher than that of ELISA(94.3%).Conclusion Compared with SAT,FPA and ELISA have better sensitivity,specificity,consistency and Kappa value,while RBT has better consistency,high sensitivity,general specificity and easy to appear false positive.By comparing and analyzing the four laboratory detection methods of brucellosis serum antibodies,fluorescence polarization method as a new technology has higher sensitivity and specificity,which is of great significance for the prevention and treatment of brucellosis.
