Objective To isolate and epidemiologically analyze the Yersinia pestis Phages collected from Marmota himalayana in Wulan County and Tongde County,Qinghai Province,and to grasp the development trend of Marmota himalayana plague epidemic and the early warning of plague prevention and control.Methods Purification was carried out by using Luria-Bertani(LB)liquid culture medium and double-layer agar plate,and the Phage lysis spectrum was analyzed.The separation rate of Phage was compared by t test.Results Fourteen and one strains of Phage were isolated from 100 Marmota himalayana intestinal samples in Wulan County and Tongde County,respectively,with the separation rate of 14.00%and 1.00%,and the difference of separation rate of Phage between the two places was statistically significant(t=3.583,P<0.001).No Phage was isolated from the paws,liver and spleen samples of the Marmota himalayana.The results of plague Phage lysis spectrum showed that 1 strain of Phage isolated from Wulan County could only lyze 18 strains of Yersinia pestis,with the lysis rate of 32.73%,while the other 13 strains and 1 strain of Phage isolated from Tongde County could lyze 55 strains of selected Yersinia pestis,with the lysis rate of 89.09%to 100.00%.Conclusion The isolation rate of Yersinia pestis Phage of samples collected by Marmota himalayana in Wulan County is higher,which is in line with the high discovery rate of plague epidemics among local animals.Although Tongde County is in a long-term resting state of plague epidemics,the isolation of Yersinia pestis Phage in samples collected by Marmota himalayana may indirectly indicate the existence of plague epidemic among animals in this area.
