Objective To investigate the mechanism of silencing TLR2 genes on chronic urinary tract infection in model rats.Methods Forty Sprague Dawley(SD)rats were divided into control group,model group,negative control(NC)group,and experimental group.The experimental group and NC group were used to establish a chronic urinary tract infection model.After modeling,sh-TLR2 and sh-NC3 reagents were injected into their tail veins once a week.The control group and model group were injected with an equal amount of physiological saline,and the body temperature of the rats was compared for three consecutive weeks.The expression level of toll like receptor 2(TLR2)in rat bladder tissue,the bacterial count in rat bladder and rat kidney,as well as the urine white blood cell count and lymphocyte count were detected.The peripheral blood inflammatory factors were also detected.In addition,the transforming growth factor(TGF)-β,level of SMAD family member 3(Smad3)mRNA expression level and p-Smad3 protein expression were detected.Results Compared with the control group,the body temperature of rats in the model group,the NC group and the experimental group significantly increased,while the body temperature of the experimental group rats was significantly lower than that of the model group and NC group.Compared with the control group,the bacterial count in the bladder and kidney tissues of the model group and NC group rats increased significantly,while the above indicators of the experimental group rats reduced markedly compared to the model group and NC group(model group:t=-3.616,-22.972,P=0.002,<0.001;NC group:t=-3.950,23.974,P=0.001,<0.001);compared with the control group,the urine white blood cell count and lymphocyte count of the model group and NC group rats were noticeably increased,with the experimental group lower than the model group and NC group(model group:t=-1.437,-1.082,P=0.168,0.293;NC group:t=-1.376,-0.712,P=0.186,0.485).Compared with the control group,the inflammatory factors in the peripheral blood of the model group and NC group rats significantly increased;compared with the NC group and model group,the inflammatory factors in the peripheral blood of the experimental group rats were substantially reduced.Compared with the control group,TGF-β and Smad3 mRNA expression levels in rats of the model group and the NC group increased;TGF-β and Smad3 mRNA expression levels were significantly reduced in rats in the test group compared with the model and NC groups(TGF-β:t=-6.371,-7.500,P<0.001;Smad3:t=-8.132,-7.406,P<0.001;p-Smad3:t=-20.586,-16.956,P<0.001).Compared with the control group,the expression levels of TGF-β,Smad3 and p-Smad3 of the model group and NC group rats increased;compared with the model group and NC group,the TGF-β,Smad3 and p-Smad3 of the experimental group rats were significantly reduced(TGF-β:t=-13.483,-23.813,P<0.001;Smad3:t=-24.575,-12.622,P<0.001;p-Smad3:t=-31.328,-16.005,P<0.001).Conclusion Silencing TLR2 can inhibit the expression of inflammatory factors in chronic urinary tract infection rats,and it can reduce the bacterial count in kidney and bladder tissues.Its mechanism may be related to the regulation of the TGF-β/SMADS signaling pathway is related.
