Yersinia pestis phage is widely distributed in nature as a bacterial bacterial of Yersinia pestis.The deep understanding of Yersinia pestis phage will play an important role in plague identification and diagnosis,disease prevention,treatment,ecological studies,and microbial evolution.In order to obtain more Yersinia pestis phage samples,the standardized collection,pretreatment,and preservation of Yersinia pestis phage samples from different bio-environmental factors in the natural foci of plague have an important impact on the quality of the samples,and the optimization factors of Yersinia pestis phage isolation and culture play an essential role in the improvement of the isolation rate of the number and species of Yersinia pestis phages in nature.The paper summarizes the isolation technology of wild-type Yersinia pestis phage including the selection of wild-type Yersinia pestis phage collection sites,the preservation conditions of the samples,methods of phage isolation and their precautions.
