硫酸铵在蛋白质含量测定中的参比作用初探

摘要
目的:中国药典2010版附录Ⅶ D 氮测定法第二法,是蛋白质含量测定的经典方法。但参考蛋白难于获得,检测结果缺乏参比性。本文尝试以分析纯硫酸铵为参考品,检测关节软骨再生支架样品中的蛋白质含量。方法分别称取三份硫酸铵,与蛋白样品一起,在相同条件下测试其回收率,用以参照评价蛋白样品的检测结果。结果硫酸铵的回收率接近100%,不考虑消化样品的差异,可以确认样品测定结果。结论在没有标准蛋白作参比的情况下,在凯氏定氮法中引入硫酸铵,可以很好的监测实验结果。
Abstract
Objective To determinate the concentration of protein,the second method of nitrogen measurement in appendix Ⅶ D of Chinese pharmacopoeia 2010 edition is always used. The complicated experiments used to leads puzzling result,for there is not right reference protein.We tried to find a way to determinate the concentration of protein in regeneration scaf old of ar-ticular cartilage by using ammonium sulfate.Methods triplet ammonium sulfate taken was used as reference to determinate the concentration of protein in the samples.Results we got a nearly 100% recovery rate of ammonium sulfate irrespective of the dif erence digestion with protein samples.Conclusions In the case of get ing no protein reference,Ammonium sulfate could be used as reference in the kjeldahl determination to improve the credibility.
