Mechanism of Shumaijiaonang on the Inflammation of the Iliac Artery after Balloon Injury in Rats
Objective To investigate the mechanism of Shumaijiaonang in inhibiting the inflammation of iliac artery after balloon in-jury in rats.Methods Thirty rats were randomly divided into sham operation group,model group,Shumaijiaonang low-dose group,Shumaijiaonang medium-dose group and Shumaijiaonang high-dose group.Percutaneous transluminal coronary angioplasty(PTCA)was performed in the model group and in the Shumaijiaonang low,medium and high-dose groups.The lower limb artery restenosis model was established after the left iliac artery was damaged by the left femoral artery.There was no balloon injury in the sham operation group.On the first day after surgery,sham operation group and model group were given normal saline instillation,1ml of normal saline per 300g body weight of rats once a day,and Shumaijiaonang low-dose and Shumaijiaonang medium-dose groups were given 0.0324g/ml,0.0648g/ml and 0.1296g/ml Shumaijiaonang saline solution instillation,respectively.The hyperplasia of left iliac artery was observed 14days after operation with elastic fiber staining,and the intima area,media area and the ratio of the intima area were measured and calculated.En-zyme-linked immunosorbent assay(ELISA)was used to detect the change of serum interleukin-6(IL-6)level.The expression of IL-6 and NF-κB in vascular wall was detected by immunohistochemistry.The protein expression of NF-κB in left iliac artery was de-tected by Western blot.Results Compared with sham operation group,the vascular lumen in model group was significantly narrowed(P<0.0001),the level of IL-6 was significantly increased(P<0.0001),the level of NF-κB was significantly increased(P<0.001).Compared with model group,the content of NF-κB protein in Shumaijiaonang medium and high-dose groups was decreased(P<0.05).Compared with Shumaijiaonang low-dose group,the level of IL-6 in Shumaijiaonang medium and high-dose groups was decreased(P<0.05).Conclusion Shumaijiaonang may inhibit vascular inflammation by intervening NF-κB pathway,thus reducing the hyperplasia of vascular intima after interventionization.
