Association of Intercellular Adhesion Molecule-1 Gene Polymorphisms with Susceptibility and Prognosis of Ischemic Cardiomyopathy
Objective To investigate the relationship between intercellular adhesion molecule-1(ICAM-1)gene polymorphism and susceptibility and prognosis of ischemic cardiomyopathy(ICM).Methods Sixty-four patients admitted for ICM were selected as the ICM group,and 138healthy subjects were selected as the control group.Blood specimens were collected and DNA was extracted from both groups.SNaPshot genotyping technique was applied to detect the SNP of ICAM-1 gene in six loci:rs12462944,rs281430,rs281434,rs3093030,rs5491,rs923366,and to analyze their correlation with the susceptibility and prognosis of ICM.Results The fre-quency of the rs5491 locus variant genotype AT was higher in the ICM group than that in the control group(P<0.05),the frequency of the allele T was higher than in the control group(P<0.05);in the dominant inheritance model and the frequency of the rs281434 locus variant genotype(AG+GG)was higher than that in the control group(P<0.05).Multivariate Logistic regression results showed that the risk of ICM in carriers of the AT type at locus rs5491 was 3.428 times higher than that of the AA type(95%CI:1.152-7.747).Multi-factorial COX regression results showed that the rs5491 locus AT genotype was a risk factor for cardiac death in ICM(HR=5.075,P<0.05),and Kaplan-Meier survival curves analysis showed that patients with ICM with the AT genotype had a higher risk of death at 3 years compared to the A A genotype(P<0.05).Conclusion Variants in ICAM-1rs5491 genotype increase the risk of ICM and patients with AT type ICM have a worse prognosis than AA type.The ICAM-1rs281434 polymorphism may be associated with sus-ceptibility to ICM.
