Neuroprotective Effect of Buyang Zhuangtong Yin on Cerebral Ischemia-reperfusion in Rats and Its Possible Mechanism
Objective To explore the neuroprotective effect of Buyang Zhuangtong Yin on cerebral ischemia-reperfusion injury(CIRI)in rats,and explore its mechanism based on Nrf2signaling pathway.Methods The SD rats were divided into blank group,sham operation group,model group,Buyang Zhuangtong Yin group,aspirin positive control group and Buyang Zhuangtong Yin combined with aspirin group,and administered by gavage daily.The rats model of middle cerebral artery occlusion/reperfusion(MCAO/R)was estab-lished by thread occlusion method.The neurological deficit score was recorded every day.Except the blank group,after reaching the time point of 1,3 and 7days,the rate of weight change were measured,the volume of cerebral infarction was observed by TTC staining,the pathological changes of brain tissue were observed by HE staining,and the expression of Nrf2 and HO-1 protein was detected by immu-nofluorescence and Western blot.Results Compared with the model group,the weight loss of rats in the Buyang Zhuangtong Yin group was reduced(P<0.05),and after 3days of administration,the ZeaLonga score was significantly decreased(P<0.05),the degree of nerve cell edema was reduced,and the necrotic nerve cells was decreased.Buyang Zhuangtong Yin could significantly up-regulated the protein expression of Nrf2 and HO-1(P<0.05)and promote the nuclear translocation of Nrf2.Conclusion Buyang Zhuangtong Yin can alleviate the nerve injury after CIRI in MCAO/R model rats,and its effect may be realized through the anti-oxidative stress pathway mediated by Nrf2/HO-1.
