摘要
目的 研究 CAPN3 在卵巢浆液性囊腺癌中的表达水平,明确其与卵巢浆液性囊腺癌预后及免疫浸润的关系.方法 通过 GEPIA2、TIMER、TISIDB、Kaplan-Meier 曲线分析 CAPN3 在卵巢浆液性囊腺癌中的表达谱、免疫浸润和预后价值.接着,利用 STRING数据库构建了涉及CAPN3 的蛋白质相互作用(protein-protein interaction,PPI)网络,GEPIA数据库分析了 10个基因与 CAPN3 的相关性.结果 CAPN3 在卵巢浆液性囊腺癌患者中呈低表达,随着卵巢浆液性囊腺癌的恶化,CAPN3 的表达量降低.CAPN3 表达水平与卵巢浆液性囊腺癌中大多数免疫标志物呈显著相关,包括 CD16、STAT5A、STAT3、LAG-3 等.此外,CAPN3 还与 4 种免疫抑制剂(BTLA、CD274、CSF1R 和 LGALS9)和 7 种免疫刺激剂(CD276、CXCL12、HHLA2、IL-6、TMEM173、TNFSF13 和 ULBP1)显著相关.高表达 CAPN3mRNA 水平在卵巢浆液性囊腺癌患者中显示出较好的总生存期,但在无进展生存期表现不明显.调控网络的分析表明,TTN与 CAPN3 相关性最为显著.结论 CAPN3 基因可能是预测卵巢浆液性囊腺癌的发生、发展及预后的肿瘤标志物.
Abstract
Objective To study the expression level of CAPN3 in ovarian serous cystadenocarcinoma,and to clarify its relationship with the prognosis and immune infiltration of ovarian serous cystadenocarcinoma.Methods GEPIA2,TIMER,TISIDB and Kaplan-Meier plotter were used to analyze the expression profile,immune infiltration and prognostic value of CAPN3 in ovarian serous cystadeno-carcinoma.Then,the STRING database was used to construct the protein-protein interaction(PPI)network involving CAPN3,and the GEPIA database was used to analyze the correlation between 10genes and CAPN3.Results The expression of CAPN3 was low in patients with ovarian serous cystadenocarcinoma.With the deterioration of ovarian serous cystadenocarcinoma,the expression of CAPN3 was de-creased.The expression level of CAPN3 was significantly correlated with most of the immune markers in ovarian serous cystadenocarcino-ma,including CD16,STAT5A,STAT3,LAG-3 and so on.In addition,CAPN3 was significantly correlated with four immunosuppressa-nts(BTLA,CD274,CSF1R and LGALS9)and seven immunostimulators(CD276,CXCL12,HHLA2,IL-6,TMEM173,TNFSF13 and ULBP1).High expression of CAPN3 mRNA in patients with ovarian serous cystadenocarcinoma showed better overall survival,but not significantly in progression-free survival.The analysis of the regulatory network showed that TTN was most significantly correlated with CAPN3.Conclusion CAPN3 gene may be a tumor marker for predicting the occurrence,development and prognosis of ovarian se-rous cystadenocarcinoma.
