Expression of Cyclophilin A and X-box Binding Protein 1 in Peripheral Blood and Arterial Tissue of Diabetes Mellitus in Rats
Objective To investigate the expression of cyclophilin A(CyPA)and X-box binding protein 1(XBP1)of type 2 dia-betes mellitus in rats.Methods SD rat models of type 1 diabetes mellitus and type 2 diabetes mellitus were established,and rats with normal diet and high fat diet were fed at the same time.The expression of CyPA and XBP1 in peripheral blood of rats in the four groups was detected by enzyme-linked immunosorbent assay(ELISA),the morphological changes of coronary artery in the four groups were ob-served by hematoxylin-eosin staining(HE staining),and the expressions of CyPA and XBP1 in femoral artery of lower limbs of rats in the four groups were detected by immunohistochemistry.Results The expression level of CyPA in peripheral blood of rats with type 2 dia-betes mellitus was higher than that in normal diet group,high fat feeding group and type 1 diabetes mellitus group,and the difference was statistically significant(t were-7.626,3.677,5.040,P<0.01).The expression level of XBP1 in peripheral blood of rats in type 2 diabetes mellitus group was lower than that in normal diet group and high fat feeding group,and the difference was statistically significant(t were 4.208,2.997;P were 0.001,0.013).The expression level of XBP1 in peripheral blood of rats in type 2 diabetes mellitus group was lower than that in type 1 diabetes mellitus group,and the difference was not statistically significant(t=1.843,P=0.084).HE stai-ning did not show significant fatty changes in the coronary arteries of the four groups.Immunohistochemistry showed that the expression level of CyPA was the highest in the arterial tissues of rats with type 2 diabetes mellitus group,while the expression level of XBP1 was the highest in the arterial tissues of rats with high fat feeding group and the lowest in the arterial tissues of rats with type 2 diabetes mellitus group.Conclusion High glucose and high fat was correlated with the expression levels of CyPA and XBP1 in in peripheral blood and ar-terial tissue of rats,and the combined effect of high glucose and high fat was stronger than that of high glucose or high fat alone.
