UPLC-MS/MS法测定苯磺酸氯吡格雷中5种遗传毒性杂质

利用超高效液相色谱-三重四级杆质谱联用仪(ultra performance liquid chromatography-tandem mass spectrometry,UPLC-MS/MS)建立了一种测定苯磺酸氯吡格雷中5种苯磺酸酯类遗传毒性杂质的方法,并进行了方法学验证.结果表明,方法的定量限和检出限均较低,其中,对硝基苯磺酰氯、对硝基苯磺酸的检出限和定量限分别为0.045 ng/mL和0.150 ng/mL,对硝基苯磺酸甲酯、对硝基苯磺酸乙酯、对硝基苯磺酸异丙酯的检出限和定量限分别为0.113 ng/mL和0.375 ng/mL.5种遗传毒性杂质在其线性范围内线性关系良好,相关系数在0.998 7~0.999 8之间,重复性RSD值在0.69%~1.85%之间,加标回收率在82.98%~119.92%之间,方法的线性、精密度、准确度、耐用性等均满足定量分析要求.因此,该方法适用于苯磺酸氯吡格雷中5种遗传毒性杂质的检测.
Determination of five genotoxic impurities in clopidogrel besilate by UPLC-MS/MS
A method for the determination of five genotoxic impurities of benzenesulfonates in clopidogrel besilate was developed using ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)and comprehensive validation of the method was carried out.The results showed that the limits of quantification(LOQ)and limits of detection(LOD)of the method were low,with the LODs and LODs for p-nitrobenzenesulfonyl chloride(PNBSC)and p-nitrobenzenesulfonic acid(PNBA)being 0.045 ng/mL and 0.150 ng/mL,respectively,and those for p-nitrobenzenesulfonic acid(PNBAS)methyl ester(MES),p-nitrobenzenesulfonic acid(PNBSE)ethyl ester(EES)and p-nitrobenzenesulfonic acid(PNBS)isopropyl ester(IPE)being 0.113 ng/mL and 0.375 ng/mL,respectively.The five genotoxic impurities showed good linearity in their linear ranges with the correlation coefficients between 0.998 7 and 0.999 8,the RSD of the reproducibility ranged from 0.69%to 1.85%,and the recoveries of the spiked standards ranged from 82.98%to 119.92%.The method linearity,precision,accuracy,and durability met the requirements for quantitative analysis.Therefore,it is suitable for the determination of five genotoxic impurities in clopidogrel besilate.
