异紫草酸纯化提取工艺及抗氧化活性研究

探究丹参多酚酸中异紫草酸的最佳分离纯化方法及其体外抗氧化活性.通过MCI Gel色谱柱、LH-20凝胶色谱柱、动态轴向加压制备液相联用分离得到高纯度异紫草酸,并与传统硅胶柱层析法分离效果进行对比.通过·OH清除实验、DPPH自由基清除实验和铁氰化钾还原实验测定异紫草酸的体外抗氧化活性.所建立方法分离得到的异紫草酸纯品归一化法百分含量达到98.7%,传统硅胶柱层析法无法分离得到异紫草酸纯品.异紫草酸对·OH和DPPH的半数清除率浓度(IC50)分别为0.277 mg·mL-1、17.43 μg·mL1,其抗氧化能力与浓度剂量呈现依赖关系.MCI-Gel/LH-20凝胶/C18键合填料柱层析法可用于高纯度异紫草酸的分离.异紫草酸具有清除自由基及抗氧化活性,可用于治疗心脑血管疾病药物的开发.
Isolation,purification and antioxidant activity of isolithospermic acid
To optimize the separation and purification technology of isolithospermic acid from extract of salvia polyphenolic acids,and study the antioxidant activity in vitro.The high purity isolithospermic acid was isolated by MCI Gel column,LH-20 Gel column,dynamic axial pressure preparation liquid chromatography,and the result was compared with those of silica gel column chromatography.The antioxidant activity of isolithospermic acid in vitro was analyzed by OH scavenging test,DPPH radical scavenging test and reducting power.The content of isolithospermic acid was 98.7%by normalization method,and the silica gel column chromatography can not separate it completely.The IC50 of isolithospermic acid to OH and DPPH were 0.277 mg·mL-1 and 17.43 μg·mL-1,the antioxidant activity was dose-dependent.The method which combined with MCI-Gel、LH-20 gel and C18 bonded packing column chromatography can be used for the separation of isolithospermic acid,and the isolithospermic acid had good antioxidant activity in vitro,it could be used for the development of cerebrovascular disease drugs.
