Construction of SSR Fingerprint on 36 Chinese Jujube Cultivars
In this study,36 Chinese jujube cultivars were analyzed by capillary electrophoresis using fluorescent M13 multi-state detection method with 12 pairs of newly developed SSR primers. Totally,99 polymorphic alleles were revealed,with an average of 8.25 for each primer pairs. Polymorphism information content values for the primer pairs ranged from 0.62–0.85,with an average of 0.75. A strategy of combining primer pair with distinct alleles for fingerprint construction was developed and applied to the 36 cultivars. The genetic similarities among cultivars range from 0.6667 to 0.9444,on which a phenetic tree showing the relationship among the cultivars was constructed. The present results provide valuable tools for the cultivar classification,germplasm identification and molecular breeding of Chinese jujube.
