Study on the expression of MAD2L1 in colorectal cancer and its relationship with immune invasion
Objective To explore the expression of Mitotic Arrest Deficient 2 Like 1(MAD2L1)in colorectal cancer tissues and its relationship with immune infiltration.Methods Based on the TCGA database,the expression difference of MAD2L1 in colorectal cancer tissues and normal colorectal tissues was analyzed,survival analysis was used to evaluate the relationship between MAD2L1 expression level and patient prognosis,and clinical correlation analysis was used to study the correlation between colorectal cancer and clinical data.The Timer database gene module was used to analyze the relationship between MAD2L1 and immune cell groups.The correlation module was used to evaluate the correlation with the level of immune checkpoints.The correlation module was used for the correlation analysis of immune cell infiltration,and the relationship between the expression of MAD2L1 in colorectal cancer tissues and immune cell infiltration was analyzed.Results Compared with normal tissues,the expression of MAD2L1 in colorectal cancer tissues was upregulated;compared with the low-expression group of colorectal cancer patients,the proportion of N-stage patients in the MAD2L1 high-expression group was different,and the difference was statistically significant(P<0.05).Prognostic analysis showed that the survival period and progression-free survival period of patients in the MAD2L1 high-expression group were higher than those of patients in the low-expression group,and the difference was statistically significant(P<0.05).Multivariate COX regression analysis showed that age and T stage can be used as independent prognostic factors affecting the prognosis of colorectal cancer patients(P<0.05).MAD2L1 was positively correlated with the infiltration level of CD8+T cells(colorectal cancer:r=0.315,P<0.001;rectal cancer r=0.352,P<0.001)and neutrophils(colorectal cancer:r=0.195,P<0.001;rectal cancer:r=0.278,P<0.001)in colorectal cancer,and negatively correlated with the infiltration level of CD4+T cells(colorectal cancer:r=-0.174,P<0.001;rectal cancer:r=-0.381,P<0.001).It was also positively correlated with the level of B cells in colorectal cancer(r=0.125,P<0.05).In colorectal cancer,MAD2L1 was positively correlated with PD-L1(r=0.159,P<0.05).In rectal cancer,MAD2L1 was negatively correlated with PD-1(r=-0.241,P<0.05).Conclusion MAD2L1 is highly expressed in colorectal cancer,and is related to the good prognosis of colorectal cancer and tumor immune infiltration,and may be a potential therapeutic target for colorectal cancer patients.
