Study on the Influencing Factors of Immunohistochemical HER-2(++)Amplification Status in Breast Cancer and Its Relationship with Clinical Pathological Characteristics
Objective To explore the influencing factors of immunohistochemistry HER-2(++)amplification status in breast cancer and its significance in clinical pathology.Methods 131 patients with invasive ductal carcinoma of the breast who were diagnosed in Luoyang Central Hospital from February 2021 to January 2023 and whose HER-2 protein expression was(++)by immunohistochemistry Envision two-step method were selected as the research subjects.All patients underwent fluorescence in situ hybridization(FISH)to determine their HER-2 gene amplification status,and the influencing factors of FISH detection of HER-2 results were analyzed.Results In the FISH detection of 131 cases of HER-2 protein(++)breast cancer,29 cases of HER-2 gene amplification(22.14%),mainly in the form of cluster amplification,accounting for 79.31%(23/29).Univariate analysis found that HER-2 gene amplification was related to the expression status of estrogen receptor(ER)and progesterone receptor(PR)(P<0.05),but it was not related to patient age,pathological type,pathological histological grading,tumor diameter,whether there was lymph node metastasis,whether there was vascular infiltration,and whether there was nerve infiltration(P>0.05).Multivariate Logistics regression analysis showed that ER-,PR-、ER+,PR-and ER-,PR+were independent predictive factors for FISH detection of HER-2 amplification(P<0.05).Conclusion By detecting the expression of ER and PR,the amplification of HER-2 gene can be effectively predicted.Further combined with FISH detection will improve the accuracy of HER-2 detection.
