The effect of ergosterol peroxide isolated from Amanita caesarea on the proliferation of ovarian cancer cells A2780S
This research aimed to study the proliferation of ovarian cancer cells A2780S induced by ergosterol peroxide isolated from Amanita caesarea.A2780S cells on the logarithmic growth phase were treated by ergosterol peroxide at the doses of 1.10,2.19,4.38,8.75,17.5 and 35 μmol/dL for 24,48 and 72 h,and cell proliferation was determined by CCK-8 assay.After treatment at dose of 2.19,4.38 and 8.75 μmol/dL for 24 h,the cell morphology was observed by inverted microscope and cell nuclear morphology was observed by DAPI assay.The early apoptosis rate of cells was determined by Annexin WPI doublestaining flow cytometry and the cell cycle was determined by PI cytometry.Ergosterol peroxide was found to inhibit the proliferation of A2780S.It increased the exfoliative and necrotic cells,and significantly increased the proportion of cells in S phase and early apoptosis of cells.The inhibition of A2780S cell proliferation by ergosterol peroxide may be through cell cycle blockage and cell apoptosis induction.
