Establishment and Application of RT-PCR Diagnostic Method for Mycoplasma Pneumonia of Swine
Objective:To establish a RT-PCR diagnostic method for Mycoplasma pneumonia of swine.Method:Sixty pigs suffering from Mycoplasma pneumonia of swine disease in Dali,Yunnan Province were selected as the research subjects.Primers and probes were designed for the conserved sequence p36 gene of Mhp,and a RT-PCR detection reaction system for Mycoplasma pneumonia of swine was established.The sensitivity,specificity,and repeatability were analyzed and clinical sample validation was conducted.Result:The sensitivity of RT-PCR detection was 102 copies,and there was no cross reaction with PRRSV,PPV,FMDV,PCV,CSFV,APP,etc.The coefficient of variation within and between batches was less than 2.00%.Positive cases were detected in 26 lung tissue samples(86.67%)and 19 nasal swab samples(63.33%).Conclusion:This study has successfully established a RT-PCR diagnostic method for Mycoplasma pneumonia of swine,which has high detection sensitivity and specificity,and good repeatability.This provides experimental basis for the accurate clinical diagnosis and scientific disease prevention and control of MPS in the later stage.
