Isolation and Identification of the Pathogen of Pleural Pneumonia in Longdong Black Goats
To establish a method for isolation and identification of the pathogen of pleuropneumonia in Longdong black goats.40 suspected cases of pleuropneumonia in black goats from the Longdong area of Gansu Province were selected as the research subjects,and identified through methods such as pathogen isolation and cultivation,staining microscopy.As a result,a total of 6 typical monoclonal strains were isolated and cultured,and microscopic examination showed that the pathogenic bacteria were spherical,short rod-shaped,long rod-shaped,or multi shaped.The biochemical tests showed that except for the negative tetrazolium reduction test of MC4 strain,the results of all other biochemical tests were consistent with those of the standard strain PG3.The serum agglutination test shows that diseased sheep serum and positive serum can undergo agglutination reaction with antigens.Six isolated pathogenic bacteria were detected by PCR and all amplified with a specific electrophoresis band of 423 bp in size.This study successfully established a method for isolation and identification of pleuropneumonia in Longdong black goats,which provides a reference basis for controlling the spread and precise prevention of this disease.
