Isolation and Identification of Staphylococcus Aureus from Clinical Mastitis in Dairy Cows in Tangshan Area
Objective:To isolate and identify Staphylococcus aureus from clinical mastitis of cows in Tangshan area,Hebei Province,and to understand its drug sensitivity.Method:The 60 cases of clinical mastitis related Staphylococcus aureus disease in a large-scale dairy farm in Tangshan,Hebei Province were selected as clinical samples for pathogen isolation and identification,Gram staining microscopy,PCR amplification identification,and drug sensitivity testing.Result:The results showed that two types of Staphylococcus aureus,Staphylococcus aureus and Staphylococcus aureus chromogene,were co isolated and cultured,and they were Gram positive.PCR detection can amplify a specific band with a size of 1459 bp.The sucrose,maltose,N-acetylglucosamine,mannose,fructose,mannitol,urea,arginine double hydrolysis,and nitrate reduction tests of the two strains of Staphylococcus isolated and cultured were positive.Staphylococcus aureus is sensitive to five antibiotics,including clindamycin,erythromycin,ofloxacin,cefotaxime,and tetracycline.Except for erythromycin,all 11 other antibiotics are sensitive to Staphylococcus aureus chromogenes.Conclusion:This study lays the foundation for improving the therapeutic effect of clinical mastitis associated Staphylococcus aureus in cows,and also provides reference value for the scientific prevention and treatment of mastitis in local cows in the future.
