组培和大棚培育下肇庆地区金线莲总黄酮含量分析

基于乙醇提取法和正交试验分析组织培养和大棚培育2种种植方式下肇庆金线莲总黄酮提取工艺,比较不同生长周期下金线莲总黄酮的含量变化.采用乙醇提取法,考察乙醇浓度、液料比和提取温度对不同种植方式金线莲总黄酮得率的影响.随后通过正交试验进行分析最优的提取工艺,验证后使用此条件测定不同月龄金线莲总黄酮含量.金线莲总黄酮最优组合为料液比1∶30 g/mL、水浴温度70℃、乙醇浓度80%,验证试验总黄酮得率为1.926%(RSD=1.11%,R2=0.992).金线莲总黄酮含量与月龄有关,随着月龄的增加,2种种植方式的金线莲总黄酮含量逐渐上升,7月龄时达到最高值.优化的乙醇提取工艺可在不同种植方式下的肇庆金线莲中稳定萃取总黄酮,并在7月龄时达到最大值,为金线莲活性成分分析和总黄酮质量评价提供参考.
Analysis of the Flavonoids Content of Anoectochilus roxburghii in Zhaoqing under Tissue Culture and Greenhouse Cultivation
To analyze the extraction technology of total flavonoids from Zhaoqing Anoectochilus roxburghii under tissue culture and greenhouse cultivation based on ethanol extraction method and orthogonal experiment,and to compare the changes in the total flavonoids content of Anoectochilus roxburghii under different growth cycles.The ethanol extraction method was used to examine the effects of ethanol concentration,liquid-to-material ratio,and extraction temperature on the Anoectochilus roxburghii flavonoid in different planting methods.Then the optimal extraction process was analyzed through orthogonal experiments,and after verification,this condition was used to determine the Anoectochilus roxburghii flavonoid of different months.The optimal combination of the Anoectochilus roxburghii flavonoid was a solid-liquid ratio of 1∶30 g/mL,a water bath temperature of 70℃,and an ethanol concentration of 80%.The yield of total flavonoids in the verification test was 1.926%(RSD=1.11%,R2=0.992).The total flavonoid content of the Anoectochilus roxburghii is related to the month.As the month increases,the total flavonoid content of Anoectochilus of the two planting methods gradually increases.And reached the highest value at the 7th month.The optimized ethanol extraction process can stably extract total flavonoids from Zhaoqing Anoectochilus roxburghii under different planting methods,and reach the maximum value in the 7th month,which provides a reference for the analysis of active ingredients of Anoectochilus roxburghii and the quality evaluation of flavonoids.
