Comparison of the Effect of Different Enzyme Digestion Methods on Extraction of Porcine Primary Hepatocytes
Porcine hepatocytes are one of the target cells of porcine circovirus.An efficient method to simply and quickly isolate hepatocytes is vital to investigate pathogenesis of porcine circovirus disease.In this article,the effects of trypsin,type II collagenase,type IV collagenase,type II collagenase and type IV collagenase mixture on extraction of porcine primary hepatocytes were compared by enzyme digestion in vitro.Results show that collagenase mixture group exhibited the most efficient cell digestion,whereas trypsin group did the opposite.Hepatocytes isolated from type II and type IV collagenase digestion represented neatly arranged shapes and clear boundaries.
