摘要

该研究以植株被咬食部位含有少量颊部细胞的唾液沉积提供的环境DNA(eDNA)可用于物种鉴定的原理为基础,借助自动提取试剂盒提取DNA和PCR程序的技术手段验证物种特异性线粒体DNA引物.试验通过片段大小和荧光团的组合来鉴定物种,采用PCR优化产物和低量DNA的聚合酶链式反应降低假阳性的可能性.结果表明,经取样数据比对后,待测样品较为精确的实现了马鹿这一物种的野外识别(>95%).

Abstract

Based on the principle that environmental DNA provided by saliva deposition containing a small amount of cheek cells in the biting sites of plants can be used for species identification,this study verified species-specific mitochondrial DNA primers by using the extraction and PCR procedures of the browser species diagnostic kit.This experiment identified the species by the combination of fragment size and fluorescent clusters,and used PCR to optimize the products and low-volume DNA polymerase chain reaction to reduce the possibility of false positives.Results shows that after sampling data comparison,the samples was accurately paired with Cervus elaphus(>95%).
