Effect of Buffalo and Holstein Milk on the Anti-inflammatory and Antioxidant Capacity of Caco-2 Cells
The aim of the experiment was to investigate the effects of buffalo milk and Holstein milk fractions on the anti-inflammatory and antioxidant capacity of Caco-2 cells.The cells were pretreated with buffalo milk and Holstein milk fractions,respectively,and then induced with LPS or H2O2 at a final concentration of 1 μmol/L.Changes in the anti-inflammatory and antioxidant capacity of the cells were detected by CCK-8,qPCR,flow cytometry,ELISA,and MDA,SOD,CAT and GSH-Px kits.The results showed that compared with the control group,the survival rate of Caco-2 cells increased significantly in all groups(P<0.05);the expression of TNF-α,IL-1β,IL-6 protein and mRNA decreased in all groups,the expression of Nrf-2 increased significantly(P<0.05),and the expression of Keap1 decreased significantly(P<0.05).After buffalo milk and Holstein milk acted on Caco-2 cells,ROS content was significantly decreased compared with the control group(P<0.05);SOD content was increased compared with the control group in all groups;MDA content was significantly decreased compared with the control group in all groups except the Holstein whole milk group and Buffalo milk whey group(P<0.05);CAT content was significantly increased compared with the control group(P<0.05);GSH-Px(GPx)content decreased significantly(P<0.05)in all groups compared to the blank group.The results showed that buffalo milk and Holstein milk could enhance the anti-inflammatory and antioxidant capacity of Caco-2 cells to different degrees after acting on the cells.
