Isolation and Identification of Beef Cattle Diarrhea Virus in Pingliang Region
To isolate and identify the source virus of diarrhea in beef cattle in Pingliang area,namely the Habitual Diarrhea Virus.Five suspected BVDV infected beef cattle feces from a large-scale beef cattle breeding farm in Pingliang,Gansu Province were collected as study materials.The virus was isolated and identified by MDBK cell inoculation,virus content determination,agar diffusion test,RT-PCR,and animal regression test.The two strains of BVDV were isolated with TCID50 values of 10-5.88/0.1 mL and 10-5.61/0.1 mL,respectively;These two viruses can be specifically amplified by RT-PCR to produce a target electrophoretic band of 804 bp in size.Agar diffusion test with BVDV Oregon C24 standard positive serum can produce a clear precipitation line,which is connected to the precipitation line formed by BVDV standard antigen;Two strains of virus isolated and inoculated into animals can exhibit typical clinical symptoms of BCVD.This study successfully isolated and identified two strains of BVDV from beef cattle farms in Pingliang area,and the pathogenicity of the strains was strong,with the same antigenicity as the standard strain,which can cause typical clinical symptoms in beef cattle.
