Study on Cellular Immune Response Induced by Endopeptidase O(PepOSS)of Streptococcus suis
Streptococcus suis(SS)is a globally significant pathogen in pig breeding and also poses a considerable zoonotic risk.In this study,a novel metal endopeptidase PepOSS was identified in SS.The amino acid sequence of PepOSS was analyzed by bioinformatics using online tools,and PepOSS protein expression and rabbit polyclonal antibody preparation were performed.The immune response of PepOSS protein to 3D4/21 cells was investigated.The results revealed that PepOSS belonged to the peptidase M13 family.The prepared polyclonal antibody exhibited reactivity towards PepOSS protein.Moreover,PepOSS protein induced a natural immune response in 3D4/21 cells,leading to significant increases in mRNA expressions of IL-6,TLR4,CXCL10,and other cytokines as well as enhanced cell phagocytosis.Additionally,it was found that the expression of pepOSS gene is regulated by the two component signaling systems VraSRSS.This study provides a solid theoretical foundation for future research related to PepOSS.
