猪链球菌肽链内切酶O(PepOSS)引起细胞免疫反应的研究

摘要
猪链球菌是全球生猪养殖业的重要病原菌,也是一种重要的人兽共患病病原.该研究在猪链球菌中鉴定了一个新的金属肽链内切酶PepOSS.利用在线工具对PepOSS氨基酸序列进行生物信息学分析,进行PepOSS蛋白表达及其兔源多克隆抗体的制备;并探究PepOSS蛋白对3D4/21细胞的免疫反应.结果显示,PepOSS属于肽酶M13家族;制备的多克隆抗体能与PepOSS蛋白产生特异性反应;PepOSS蛋白可引起3D4/21细胞天然免疫应答反应,经过PepOSS蛋白刺激后,IL-6、TLR4、CXCL10等细胞因子的mRNA表达量显著增加,且能增强细胞的吞噬作用;并证实pepOSS基因的表达受二元信号转导系统VraSRSS的调控.该研究为后续PepOSS相关研究提供了良好的理论依据.
Abstract
Streptococcus suis(SS)is a globally significant pathogen in pig breeding and also poses a considerable zoonotic risk.In this study,a novel metal endopeptidase PepOSS was identified in SS.The amino acid sequence of PepOSS was analyzed by bioinformatics using online tools,and PepOSS protein expression and rabbit polyclonal antibody preparation were performed.The immune response of PepOSS protein to 3D4/21 cells was investigated.The results revealed that PepOSS belonged to the peptidase M13 family.The prepared polyclonal antibody exhibited reactivity towards PepOSS protein.Moreover,PepOSS protein induced a natural immune response in 3D4/21 cells,leading to significant increases in mRNA expressions of IL-6,TLR4,CXCL10,and other cytokines as well as enhanced cell phagocytosis.Additionally,it was found that the expression of pepOSS gene is regulated by the two component signaling systems VraSRSS.This study provides a solid theoretical foundation for future research related to PepOSS.
