摘要

研究旨在鉴定鸡环状RNA(circRNA)circ_UBE2D2并预测其功能.通过RT-PCR和测序,确认鸡circ_UBE2D2由外显子4至8构成共374 bp的环状结构.生物信息学预测显示其具有物种保守性.在细胞核和细胞质中均有富集,在胸腺、脾脏、心脏和肝脏中高表达,而在回肠、盲肠和哈德林腺中低表达.circ_UBE2D2可能与AGO2、CELF2、PTBP1和HNRNPC等RNA结合蛋白互作,并可能靶向gga-miR-177和gga-miR-7472-5p.其靶基因可能通过Wnt信号通路、MAPK信号通路、C型凝集素受体信号通路和ErbB信号通路参与细胞生物学过程.因此,鸡circ_UBE2D2存在且参与调节细胞生物学过程.

Abstract

This study aims to characterize chicken circular RNA(circRNA)circ_UBE2D2 and predict its function.By RT-PCR and sequencing,it is confirmed that chicken circ_UBE2D2 consists of exons 4 to 8,which form a total of 374 bp of the circular structure.Bioinformatics prediction shows that it is species-con-served.It is enriched both in nucleus and cytoplasm and highly expressed in the thymus,spleen,heart and liver,while lowly expressed in ileum,cecum and Harderian gland.CFirc_UBE2D2 may interact with RNA-binding proteins such as AGO2,CELF2,PTBP1,and HNRNPC,and target gga-miR-177 and gga-miR-7472-5p.Its target genes may be involved in cell biological processes through the Wnt signaling pathway,MAPK signaling pathway,C-type lectin receptor signaling pathway and ErbB signaling pathway.In conclusion,chicken circ_UBE2D2 exists and is involved in regulating cell biological processes.
