Correlation analysis of miR-497-5p and miR-3607-3p with cervical cancer infected with high-risk HPV
Objective To explore the correlation between miR-497-5p,miR-3607-3p,and high-risk human papillomavirus(HPV)infection in cervical cancer.Methods A retrospective study was conducted on the clinical data of 82 patients who underwent cervical biopsy in the gynecology department of Suzhou Kowloon Hospital,School of Medicine,Shanghai Jiao Tong University,from July 2018 to July 2023.The patients were divided into three groups:chronic cervical inflammation(n=20),cervical intraepithelial neoplasia(CIN)(n=34),and cervical cancer(n=28).In addition,41 healthy individuals who underwent a physical examination during the same period were selected as a control group at a ratio of 2:1.The expression of miR-497-5p and miR-3607-3p in each group was compared with that in the patients with high-risk HPV infection,and the relationship between miR-497-5p and miR-3607-3p expression and high-risk HPV infection was analyzed.The expression of miR-497-5p and miR-3607-3p in patients with different clinical pathological characteristics of cervical cancer was compared.The diagnostic value of miR-497-5p and miR-3607-3p expression for cervical cancer infected with high-risk HPV was evaluated through ROC curve analysis.Results The order of miR-497-5p and miR-3607-3p expression in the four groups was cervical cancer group<CIN group<chronic cervical inflammation group<healthy control group(P<0.05),and the order of high-risk HPV infection rate in the four groups was cervical cancer group>CIN group>chronic cervical inflammation group>healthy control group(P<0.05).With the worsening of cervical disease,the incidence of HPV infection involving types 16 and 18 gradually increased,while the incidence of HPV infection with other types gradually decreased(<0.05).The expression of miR-497-5p and miR-3607-3p was negatively correlated with high-risk HPV infection(r--0.793,-0.697,P<0.05).There were statistically significant differences(P<0.05)in the expression of miR-497-5p and miR-3607-3p among cervical cancer patients with different FIGO stages,vascular or parametrial metastases,and lymph node metastases.ROC curve analysis showed that the area under the curve(AUC)scores of miR-497-5p and miR-3607-3p expression for the diagnosis of cervical cancer infected with high-risk HPV were 0.829 and 0.807,respectively.The diagnostic sensitivity and specificity for miR-497-5p were 92.9%and 83.3%,and for miR-3607-3p were 100.0%and 64.8%,respectively,indicating a certain diagnostic value.The AUC of two joint diagnoses of cervical cancer with high-risk HPV was 0.907,with a sensitivity of 100.0%and a specificity of 77.8%.Conclusions miR-497-5p and miR-3607-3p are expressed at low levels in cervical cancer,and are closely related to high-risk HPV infection and clinical pathological characteristics of cervical cancer.They can be used as effective indicators for the diagnosis of cervical cancer with high-risk HPV infection,and their combined diagnostic value is higher.
