目的 对1例猴痘病毒(MPXV)感染者疱疹液拭子样本进行全基因组测序分析,为MPXV感染疫情防控提供参考依据.方法 对2023年7月浙江省温州市1例MPXV感染者疱疹液拭子样本使用第三代测序技术(TGS)进行全基因组测序,对得到的病毒序列进行同源性分析并构建进化树,分析变异位点等.结果 该MPXV全基因组序列与参考株NC-063383.1相比相似性为99.92%,共有87个核苷酸位点发生变异、43个核苷酸位点删除并最终导致36个氨基酸变化.该序列在进化树上位于Ⅱb亚型进化分支,与中国其他地区通报的猴痘病毒分型一致.结论 通过三代测序确定了一株属于Ⅱb进化分支C.1(B.1.3.1)谱系的MPXV,溯源分析显示其与同时期葡萄牙暴发的毒株位于同一个进化分支上.但该分支突变带来的影响尚不明确,仍需进一步研究.
Whole genome sequence analysis of a mpox virus strain of clade Ⅱb
Objective To perform whole-genome sequencing of a vesicular fluid swab sample from a case of mpox virus(MPXV)infection to provide insights into MPXV outbreak prevention and control.Methods Herpes fluid swab samples from a case of MPXV infection in Wenzhou,Zhejiang Province in July 2023 were subjected to third-generation sequencing(TGS)for whole-genome analysis.The obtained viral sequences were compared using homology analysis,an evolutionary tree was constructed,and the mutation sites were examined.Results The MPXV genome sequence exhibited 99.92%similarity with the reference strain NC-063383.1,with 87 nucleotide mutations and 43 nucleotide deletions,resulting in 36 amino acid changes.Phylogenetic analysis placed this sequence within the clade Ⅱb evolutionary branch,and this is consistent with MPXV genotyping reported in other regions of China.Conclusions Third-generation sequencing identified an MPXV strain belonging to the evolutionary lineage C.1(B.1.3.1)within clade Ⅱb.Phylogenetic analysis traced this strain to the same evolutionary branch as that of a concurrent outbreak in Portugal.However,the implications of the mutations in this branch remain unclear and require further investigation.
