Whole genome sequence analysis of a mpox virus strain of clade Ⅱb
Objective To perform whole-genome sequencing of a vesicular fluid swab sample from a case of mpox virus(MPXV)infection to provide insights into MPXV outbreak prevention and control.Methods Herpes fluid swab samples from a case of MPXV infection in Wenzhou,Zhejiang Province in July 2023 were subjected to third-generation sequencing(TGS)for whole-genome analysis.The obtained viral sequences were compared using homology analysis,an evolutionary tree was constructed,and the mutation sites were examined.Results The MPXV genome sequence exhibited 99.92%similarity with the reference strain NC-063383.1,with 87 nucleotide mutations and 43 nucleotide deletions,resulting in 36 amino acid changes.Phylogenetic analysis placed this sequence within the clade Ⅱb evolutionary branch,and this is consistent with MPXV genotyping reported in other regions of China.Conclusions Third-generation sequencing identified an MPXV strain belonging to the evolutionary lineage C.1(B.1.3.1)within clade Ⅱb.Phylogenetic analysis traced this strain to the same evolutionary branch as that of a concurrent outbreak in Portugal.However,the implications of the mutations in this branch remain unclear and require further investigation.
