Pathogen analysis and genetic characteristics of VP1 gene of coxsackievirus B2 in hand,foot and mouth disease cases in two areas of Yunnan province in 2021
Objective To characterize the pathogens of hand,foot and mouth disease(HFMD)in the Qujing city and Wenshan prefecture of Yunnan province in 2021 and analyze characteristics of VP1 genes of 6 detected coxsackievirus B2(CVB2)and to provide molecular epidemiological data for future prevention and control of HFMD and CVB2.Methods Fecal samples of HFMD cases in Qujing city and Wenshan prefectures from January to December 2021 were collected and detected for enteroviruses by real-time quantitative reverse transcription PCR(real-time qRT-PCR),and VP1 genes of other enteroviruses identified(non-EV-A71 and non-CVA16)were sequenced.The VP1 gene of CVB2 reference sequences downloaded from the GenBank and the genetic characteristics were analyzed by Mega 5.2 software.Phylogenetic trees were constructed by the neighbor-joining method in the Kimura 2-parameter substitution model.Results In 2021,1 164 fecal specimens of HFMD cases were collected from the two regions in Yunnan province.After real-time qRT-PCR testing,927 positive specimens were detected(positive rate 79.64%,927/1 164),including 109 EV-A71(9.36%),343 CVA16(29.47%),and 475 other enteroviruses(51.24%).Gene sequencing was performed on other enteroviruses,and 6 CVB2 strains were identified(2 in Qujing and 4 in Wenshan),accounting for 1.26%(6/475).Phylogenetic analysis showed the 6 strains belonged to cluster 2 in genotype D.Conclusion In 2021,other enteroviruses are the main pathogens and CVB2 virus is detected in samples of HFMD cases in both Qujing and Wenshan of Yunnan province,which belongs to cluster 2 of genotype D.It suggests that further typing of other enteroviruses should be strengthened in order to comprehensively understand the pathogen composition of HFMD and provide a basis for scientific prevention and control.
