精索静脉曲张引起大鼠生殖功能改变及睾丸HSP90α表达变化

目的:探讨精索静脉曲张后大鼠生殖功能的变化和睾丸中热休克蛋白90α( HSP90α)的表达变化及规律。方法将32只SD大鼠随机分成4组:对照组和3个精索静脉曲张组(造模成功后第1、3、6周)。分别测定各组大鼠的相应脏器指数,检测精子相关指标、睾丸中HSP90α的表达。结果精索静脉曲张后,大鼠附睾指数在1、3周时显著低于对照组;精子活力1、3、6周组显著低于对照组,精子畸形率在1周3周时间点明显高于对照组,而1、3周时间点精子存活率显著低于对照组,其他试验组与对照组相比无显著差异;1、3、6周组睾丸中HSP90α的表达均显著高于对照组( P<0.05)。结论精索静脉曲张可引起大鼠生殖系统损伤以及睾丸中HSP90α的高表达,提示HSP90α可能保护机体在静脉曲张状态下受到的损伤。
Objective To investigate the changes of reproductive function and Hsp 90αgene expression in reproductive system of rat with varicocele .Methods Healthy 32 SD rats were randomly divided into 4 groups:control group and 3 varicocele group(1,3 and 6w after varicocele).Organ index and HSP90αgene expression in testicle were determined.Result At 1 and3w the epididymis index was significantly lower than it in the control;sperm motility rate is significantly lower than the control group at 1,3w and 6w(P<0.05),Sperm deformity and morality rate were significantly higher than the control group at 1 and 3w(P <0.05).Gene expression of HSP 90αin testicle was significantly higher than those in the control group at1,3 and 6w group(P<0.05).Concluis on Varicocele resulted in the damage of reproductive system,and the increased expression of HSP90αgene in testicle.The increased expression of HSP90αgene may protect cells from varicocele damage.
