Objective:To investigate the high-fat diet combined with low dose of streptozotocin (STZ)-induced type 2 diabetes mel itus (T2DM) rats. Methods:Male Spragye-Dawley rats were fed with high-fat diet for 6 weeks. Then streptozotocin 40 mg/kg was injected intraperitoneal y once to induce diabetic rat model. Diabetic rats were randomly divided into model group, metformin (100 mg/kg) group. And normal control group was fed with basal feed. After intragastric administration of metformin and isopyknic distil ed water daily for 4 weeks to rats of metformin group and normal control group respectively, the fasting blood glucose (FBG), fasting insulin (FINS), free fat y acids (FFA), triglyeride (TG), total cholesterol (TC) were determined, and homeostasis model assessment of insulin resistance (HOMA-IR) were calculated. Results:Compared with the normal control group, the levels of FBG, FINS, FFA, TG, HOMA-IR were significantly increased (P <0.05). The FBG, FINS and HOMA-IR were significantly decreased in the metformin group compared with those in the model group (P <0.05). Conclusion:The method of inducing T2DM rats by high-fat feeding combined with STZ (40 mg/kg) is effective and the models are stable.
