大孔吸附树脂纯化银杏黄酮的工艺研究

目的:研究大孔吸附树脂纯化银杏总黄酮的工艺条件及参数。方法:采用静态吸附、解吸,从6种大孔树脂中筛选用于银杏总黄酮分离的最佳树脂,并通过动态行为系统考察最佳大孔树脂的吸附性能和最优洗脱参数。结果:HPD-450型大孔树脂为分离银杏黄酮的最佳树脂,其分离的最佳工艺为,上样量为6.5倍柱体积药液,以5mL/min 的流速,pH 值为4.0上柱,用400mL85%乙醇以4mL/min 流速进行洗脱,结论:按照最佳工艺可获得总黄酮纯度达26%以上的银杏叶提取物。
OBJECTIVE:To study process conditions and parameters of purification of total gingko flavones-glycoides by macroporous adsorptive resin. METHODS:To select the optimal resin used to separate glycoides among 6 different macroporous resins by static adsorption and desorption and investigate the adsorption property and optimal elution parameters of the best resin by dynamic adsorption. RESUTS:The results show that HPD-450 macroporous resin is the best resin used for separating glycosides,and the optimal technological conditions were as fol ows: the sample amount 6.5 times the column volumes, sample flow velocity 5 mL/min, pH value 4.0, eluting with 400 mL of 85% ethanol at the eluting velocity of 4mL/min. CONCLUSIONS:It turned out that total flavonoids purity more than 26% of ginkgo biloba extract according to the optimum.
