The effect of ovariectomy and muscle injection of dexamethasone on rat vascular calcium content and bone mineral density

目的:观察去卵巢和地塞米松肌肉注射对大鼠血管钙含量和骨密度的影响。方法:对雌性 Wistar 大鼠分别进行去卵巢及肌肉注射地塞米松,观察去卵巢及地塞米松肌肉注射对血管钙含量和骨密度的影响。雌性Wistar大鼠共分为4组,即对照组、假手术组、去卵巢组和地米注射组,10周后,进行主动脉钙含量、碱性磷酸酶(alkaline phosphatase, ALP)活性及骨钙素(osteocalcin, OC)含量测定,判断血管钙化程度;用骨密度仪测定大鼠骨密度以反映骨质疏松程度。结果:去卵巢组和地米注射组血管钙含量均升高,明显高于对照组和假手术组(P<0.01﹚;两组腰椎骨密度均低于对照组(P<0.01,P<0.05﹚,去卵巢组股骨骨密度亦低于对照组(P<0.05﹚。结论:去卵巢和肌注地塞米松均可导致大鼠骨密度降低,同时可增加血管钙含量,其机制有待进一步探讨。
Objective:to observe the effect of ovariectomy and dexamethasone intramuscular on vascular calcium content and bone density of rat. Methods:the female Wistar rats were separately deal with the ovariectomy and muscle injection of dexamethasone, to observe the effects on vascular calcium content and bone mineral density. Female Wistar rats were divided into 4 groups, namely the control group, sham-operation group, ovariectomy group and the dexamethasone injection group, 10 weeks later, aortic calcium, alkaline phosphatase(ALP) activity and osteocalcin (OC) content were determinated, to reflect the degree of vascular calcification; Rat bone density is measured by bone mineral density instrument to reflect the degree of osteoporosis. Results:the ovariectomy group and dexamethasone injection group vascular calcium were elevated, were significantly higher than the control group and sham-operation group (P<0.01);the lumbar vertebra bone mineral density were lower than that of control group in the two groups (P<0.01, P<0.05), the femoral bone mineral density in ovariectomy group is lower than that of the control group (P<0.05). Conclusions:the ovariectomy and muscle injection of dexamethasone to rat may lead to bone mineral density reduce, at the same time can increase the blood calcium content, its mechanism remains to be further discussed.
