RP-HPLC analysis of paeoniflorin and albiflorin content in Paeonia lactiflora Pall. callus
AIM:To establish a sensitive RP-HPLC method for determining the active ingredient content of peoniflorin and albiflorin inPaeonia lactifloraPal. calus cultivated in subculture process.METHODS:The separation was performed on SinoChrom ODS-BP column (250 mm×4.6 mm, 5μm), with acetonitrile and 0.05% potassium dihydrogen phosphate solution (v/v, 11:89) as mobile phase, detected at 230 nm wavelength, and the flow rate was 1.0 mL/min.RESULTS:The standard curves of peoniflorin showed a good linearity in a certain range.CONCLUSION:The method is accurate, reliable and suitable for determination of albiflorin and paeoniflorin inPaeoniae radix Alba calus simultaneously. It lays a foundation for further condition optimization of calus culture to produce albiflorin and paeoniflorin inRadix Paeoniae Alba.
