To develop a method of safety assessment for Clove essential oil. Methods:the content of Clove essential oil was determined by GC with eugenol as a quantitative indicator and its safety was evaluated by the use of acute skin irritation test, multiple skin irritation and eye irritation test. Results:The linearity of eugenol content was good when its concentration was in the range between 0.0997~1.9940 mg/ml, the average rate of recovery was 99.01%, RSD was 1.89%(n=6). With regard to acute skin irritation test and eye irritation test, Clove essential oil was nonirritant at a concentration of 50%. In the multiple skin irritation, Clove essential oil was mild irritant at concentrations of both 75% and 100%. Conclusion:The GC method is fast, simple, accurate and suitable for quality control of Clove essential oil. Clove essential oil is mild irritant for eyes and skin of animal.
