HPLC-ICP/MS determination selenomethionine selenium protein amino acids
To establish a method for detection of selenium in selenium amino acid generation, detection and it can be used in food nutrition fortifier of selenoproteins. Solution of selenoprotein enzymes; 5mM methanol solution and ammonium citrate plus 2% as the mobile phase, ion exchange column chromatography of seleno amino acids; content of ICP/MS detection of seleno amino acids. Using proteinase K, trypsin, pronase enzyme solution with citric acid, and adjusting the pH value of the mobile phase (pH=4.9) to SeCys, SeMet seleno amino acid separation. The detection limits were:; the linear range is: Se 0~200μ g/L; recovery of standard addition: 75%~120%. Analysis of the method can be used in generation of selenoproteins in amino acid.
