高丹草新品系的SSR分析

SSR Analysis of New Strains of Sorghum-Sudangrass

对11份高丹草新品系(品种)进行了SSR多态性分析.用筛选出的9对SSR适宜引物对高丹草各材料基因组DNA进行PCR扩增,得到318个多态性位点,多态性位点比率达95.78%.引物Xtxp13及Xtxp67扩增的SSR指纹图谱差异明显,可作为11个高丹草新品系(品种)鉴别的分子依据.11份高丹草材料间的GD值变动在0.4286~0.7226之间,以GD值0.61为基准,可将11份材料分为3类:第一类包括SLCN01、SLCN02、SLCN03、SLCN09、SLCN10和蒙农青饲3号高丹草;第二类包括SLCN04、SLCN05、SLCN06、SLCN08;SLCN07单独为一类.
SSR polymorphism of 11 new strains of Sorghum-Sudangrass was analyzed. Genomic DNAs of Sorghum-Sudangrass were amplified with nine pairs of SSR primers, there were 318 polymorphic loci, the percentage of polymorphic loci reached 95. 78%. There were significant differences between fingerprint map amplified with primer Xtxpl3 and fingerprint map amplified with primer Xtxp67, which can be used as molecular basis for identifying the eleven new strains (cultivars) of Sorghum-Sudangrass. Genetic distance (GD) among the new eleven Sorghum-Sudangrass strains ranged from 0. 4286 to 0. 7226, using the GD value of 0. 61 as threshold, the eleven new strains of Sorghum-Sudangrass can be clustered into three groups: group one: SLCN01, SLCN02, SLCN03, SLCN09, SLCN10 and Mengnong qingsi No. 3; group two: SLCN04, SLCN05, SLCN06, SLCN08; group three: SLCN07.
