miR-515对凝集素-2的调控作用以及宫内膜间质细胞生物学行为的影响

摘要
目的 探讨微小 RNA-515(micro RNA-515,miR-515)对凝集素 2(Intelectin 2,ITLN2)的调控作用以及对子宫内膜间质细胞(endometrial stroma cells,ESCs)生物学行为的影响.方法 分离ESCs细胞并进行传代培养,分为对照组(转染NC)、过表达组(转染miR-155 mimics)、抑制组(转染miR-515 inhibitor)和逆转组(转染miR-515mimics+pcDNA-ITLN2),检测各组ESCs增殖、凋亡、侵袭和转移情况,以及RNA和蛋白的表达.结果 过表达组细胞增殖、迁移细胞个数和侵袭细胞个数显著升高、凋亡率显著下降(P<0.05);抑制组细胞增殖、迁移细胞个数和侵袭细胞个数值显著下降,凋亡率显著上升(P<0.05).过表达组miR-515表达显著升高,ITLN2 mRNA和ITLN2蛋白表达显著下降(P<0.05);抑制组miR-515表达显著下降,ITLN2mRNA和ITLN2蛋白表达显著上升(P<0.05);逆转组介于过表达组和抑制组之间(P<0.05).野生型ITLN2+miR-515的荧光强度显著下降(P<0.05).结论 miR-515可以负调控ITLN2的表达,促进ESCs细胞的增殖、侵袭和转移,抑制ESCs细胞的凋亡.
Abstract
Objective To investigate the regulatory effect of microRNA-515(miR-515)on lectin 2(ITLN2)and the effect on biological behavior of endometrial stromal cells(ESCs).Methods ESCs cells were isolated,subcultured and divided into control group(transfected NC),overexpression group(transfected miR-155 mimics),inhibition group(transfected miR-515 inhibitor),and reversal group(transfected miR-515 mimics+pcDNA-ITLN2).The proliferation,apoptosis,invasion,metastasis,as well as RNA and protein expression of ESCs in each group were detected.Results The overexpression group showed a significant increase in cell proliferation,migration,and invasion,while the apoptosis rate decreased significantly(P<0.05);The overexpression group showed a significant increase in miR-515 expression,while the expression of ITLN2 mRNA and ITLN2 protein decreased significantly(P<0.05);The expression of miR-515 in the inhibition group decreased significantly,while the expression of ITLN2 mRNA and ITLN2 protein increased significantly(P<0.05).The reversal group was between the overexpression group and the inhibition group(P<0.05).The fluorescence intensity of wild-type ITLN2+miR-515 decreased significantly(P<0.05).Conclusion MiR-515 can negatively regulate the expression of ITLN2,promote the proliferation,invasion and metastasis of ESCs cells,and inhibit the apoptosis of ESCs cells.
