Expression of Treg cells and related factors in placental tissue of pregnant women with severe pre-eclampsia
Objective To investigate the expression and significance of Treg cells and related factors in placental tissues of pregnant women with severe pre-eclampsia(SPE).Methods A total of 74 patients with SPE(SPE group)and 75 women with Full-term production(control group)admitted between January 2020 and January 2022 were included as study subjects.Baseline data and laboratory examination data were collected from the study subjects,and the expression levels of CD4+CD25+Treg cells in peripheral blood were detected,and the expression levels of transcription factor Foxp3 mRNA and transforming growth factor β1(TGF-β1)in peripheral blood Treg cells were detected by fluorescence quantitative polymerase chain reaction(PCR),and protein blotting(Western blot)was used to the expression levels of Foxp3 and TGF-β1 protein in placental tissues were detected.Results The percentage of serum CD4+CD25+Treg was lower in the SPE group than in the control group;the serum IL-10 and TGF-β1 levels were higher in the SPE group than in the control group(P<0.05),and the HLA-G levels were lower than in the control group(P<0.05);the expression levels of Foxp3 mRNA in serum and placental tissues were lower in the SPE group than in the control group(P<0.05).The expression levels of TGF-β1 mRNA in serum and placental tissues were higher in the SPE group than in the control group(P<0.05);the expression level of Foxp3 protein in placental tissue of SPE group was lower than that of control group,and TGF-β protein expression level was higher than that of the control group;There was a positive correlation between TGF-β1 level in peripheral blood of SPE patients and TGF-β1 protein level in placental tissue(r=0.645,P<0.05).Conclusion The expression of Treg cell-related factors(Foxp3,TGF-β1)in peripheral blood and placental tissues of SPE patients was changed.
