摘要
目的:基于网络药理学探讨宫毅教授临床治疗老年便秘核心用药的作用机制.方法:在TCMSP、Drugbank、TTD、Disgenet和GeneCards数据库中检索核心药物白术、党参、黄芪、当归、紫菀以及疾病的靶点;运用Venny 2.1获取交集靶点,继而于String中进行PPI分析;通过Cytoscape 3.7.1以及Metascape数据库完成可视化以及GO、KEGG富集分析.结果:得到41个活性成分、217个药物靶点、1079个疾病靶点以及94个交集靶点.其中41个活性成分中含有木犀草素、山奈酚和槲皮素等;217个核心靶点含有AKT1、IL6、TNF等;信号通路以癌症通路、MAPK通路、PI3K-AKT通路和AGE-RAGE通路为主.结论:宫毅教授治疗老年便秘以白术、党参、黄芪、当归和紫菀为核心药物,其可能通过多靶点、多通路协同交互发挥作用,需进一步实验验证.
Abstract
Objective To explore the mechanism of Professor GONG Yi's core medication in the treat-ment of senile constipation based on network pharmacology.Methods The core drugs Baizhu(Atractyl-odis Macrocephalae Rhizoma),Dangshen(Codonopsis Radix),Huangqi(Astragali Radix),Danggui(Angel-icae Sinensis Radix)and Ziwan(Asteris Radix Et Rhizoma)and the targets of diseases were retrieved from databases of TCMSP,Drugbank,TTD,Disgenet and GeneCards.The intersection targets were obtained by using Venny 2.l,and then PPI analysis was performed in String.The visualization and GO,KEGG enrich-ment analysis were completed by using Cytoscape 3.7.1 and Metascape database.Results Total 41 ac-tive ingredients,217 drug targets,1079 disease targets and 94 intersection targets were obtained.Luteolin,kaempferol and quercetin,etc.were included in the 41 active ingredients.AKT1,IL6 and TNF,etc.were included in the 217 core targets.The signal pathways were mainly cancer pathway,MAPK pathway,PI3K-AKT pathway and AGE-RAGE pathway.Conclusion Professor GONG Yi treats senile constipation with Baizhu,Dangshen,Huangqi,Danggui and Ziwan as the core drugs.Further experiments are needed to verify that it may play a role through multi-target and multi-channel synergistic interaction.
