Mechanism of Shengji Yuhong Paste in Promoting Wound Healing Through Expression of FGFR-1
Objective To investigate the mechanism of Shengji Yuhong Paste in promoting wound heal-ing in model rats with local skin and muscle defect of anal margin.Methods Total 40 male rats were randomly divided into blank group,model group,Meibao Shirun Shaoshang Cream(MEBO)group and Shengji Yuhong Paste group,with 10 rats in each group.Except the blank group,the other 3 groups were treated with sterilized surgical scissors to establish the model rats with local skin and muscle defect(diam-eter 1 cm)at the anal margin.After the modeling was successful,the rats in the MEBO group(1.00 g/kg)and Shengji Yuhong Paste group(2.08 g/kg)were smeared with corresponding drugs on the wound sur-face,and the rats in the model group and blank group were smeared with equal volume normal saline for 14 consecutive days.The general state and body mass of rats were observed before and after wound adminis-tration.The wound area and healing rate were calculated.The histopathologic changes of wound skin were observed by HE staining.The abundance of growth factor receptor-1(FGFR-1)expression in rat wound skin fibroblasts was detected by immunohistochemistry.Results 1)Body mass and general status:Wa-ter intake,food intake and body mass of rats in the blank group increased steadily.On the 3rd day after modeling,the body mass of rats in the modeling groups slightly decreased compared with that in the blank group(P<0.05).On the 7th and 14th day of administration,the body mass of rats in each group in-creased steadily,which was lower in each modeling group than in blank group(P<0.05).On the 3rd,7th and 14th day of administration,the body mass of each modeling group was slightly lower than that of ME-BO group and Shengji Yuhong Poste group,but the difference was not significant(P>0.05).2)Wound area and healing rate:Compared with the model group,the wound area of MEBO and Shengji Yuhong Paste groups was significantly decreased(P<0.05)and the healing rate was significantly increased(P<0.05)on the 3rd and 7th day of administration.On the 14th day,the wounds of the model group,MEBO group and Shengji Yuhong Paste group were basically healed.3)Histopathological observation of the wound skin:The epidermal structure of the blank group was intact without inflammatory cell infiltration.In the model group,small focal necrosis,inflammatory cell infiltration,epidermal thickening and spinous cell proliferation were observed in epidermis and superficial dermis.Small area of granulomatous tissue and connective tissue hyperplasia were observed in the dermis.In Shengji Yuhong Paste group and MEBO group,there were different degrees of improvement.4)FGFR-1 expression abundance in wound skin tis-sue:On the 7th day of administration,compared with the blank group,FGFR-1 expression abundance in the modeling groups was significantly increased(P<0.05).Among the modeling groups,the abundance of FGFR-1 expression in Shengji Yuhong Paste group was the highest,followed by MEBO group,and the difference between the two groups was statistically significant(P<0.05).On the 14th day of administra-tion,FGFR-1 expression abundance in the model group and MEBO group had no significant change com-pared with that on the 7th day of administration(P>0.05),but decreased in Shengji Yuhong Paste group(P<0.05).Conclusion Shengji Yuhong Paste can promote healing of skin wounds in rats,and this effect may be related to the expression abundance of FGFR-1 protein.
