摘要

骨稳态包括成骨细胞和破骨细胞介导的骨形成与骨吸收,骨组织的完整性依赖二者间的动态平衡.研究表明,骨稳态的建立除受年龄、性别、生活方式等影响外,还与遗传因素密切相关.表观基因组学描述了一种在不改变DNA序列状态下控制基因表达的遗传机制,包括DNA甲基化、组蛋白修饰和非编码RNA.而运动作为外源性刺激因素,在"遗传-环境"范畴内可通过干预基因的表观组学状态影响疾病发展.这表明在调控骨稳态过程中,运动能成为宏观与微观刺激的桥梁.基于此,本文全面综述了表观基因组学调控骨稳态的详细机制,深入探讨了运动干预表观基因组学影响骨稳态的潜在途径,并总结了"遗传-环境-骨骼"间完整的作用框架,为运动调控内源性因素干预骨稳态提供了新视角.

Abstract

Bone homeostasis involves bone formation and resorption mediated by osteoblasts and osteoclasts.The integrity of bone tissue depends on the dynamic balance between the two.Studies have shown that the establishment of bone homeostasis is closely related to genetic factors,in addition to age,gender,and lifestyle.Epigenomics describes a genetic mechanism that controls gene expression without altering the DNA sequence,including DNA methylation,histone modification,and non-coding RNAs.Exercise,as an exogenous stimulus,influences disease progression by interfering with the epigenomic state of genes in a genetic-environmental context.This suggests that exercise serves as a bridge between macro-and micro-stimuli in the regulation of bone homeostasis.Based on this,this paper comprehensively reviews the detailed mechanism of epigenomic regulation of bone homeostasis,explores the potential pathways through which exercise interferes with epigenomic effects on bone homeostasis,and summarizes the complete framework of the genetic-environmental-skeletal interactions,which provides a new perspective for the regulation of bone homeostasis by exercise and the intervention of endogenous factors.
