Bone is one of the largest organs in the body, and abnormal bone development and homeostasis can cause a variety of bone diseases, such as bone genetic diseases, degenerative diseases(such as osteoporosis, osteoarthritis)and fractures.FGFR1,a member of the FGFR family, plays important roles in skeleton development and homeostasis.Gain-of-function missense mutations in FGFR1 lead to bone genetic diseases such as craniosynostosis syndrome.FGFR1 also involved in the regulation of bone formation, bone resorption processes, and the development of osteoarthritis. In addition,FGFR1 was expressed in bone-related cells at different stages of bone injury repair, suggesting its involvement in the process of bone repair. This paper reviews the role and mechanism of FGFR1 in bone development, homeostasis maintenance and injury repair, providing new ideas for the treatment of bone diseases by interfering FGFR1 signaling.
