Effect of LY294002 on the proliferation and apoptosis of colorectal cancer RKO cells through PI3K/Akt pathway
Objective To study the effect and mechanism of LY294002 on the proliferation and apoptosis of colorectal cancer RKO cells.Methods The expression of p-Akt in colorectal cancer tissues and adjacent cancer tissues was detected by immuno-histochemistry.Human colorectal cancer RKO cells were cultured and divided into 3 groups:blank group,DMSO group and LY294002 group.Cells'proliferation were determined by CCK-8 assay to analyze the effect of LY294002 on the proliferation of human colorectal cancer RKO cells.The effect of LY294002 on the apoptosis of RKO cells was detected by flow cytometry.Western blot was used to detect the key molecules,such as p-Akt,Akt,Bax and Bcl-2.Results P-Akt was significantly up-regulated in colorectal cancer tissues compared with adjacent cancer tissues.The results of CCK-8 showed that compared with the other two groups,the number of viable cells in LY294002 group was decreased(P<0.05)and the apoptosis rate of LY294002 group was increased significantly(P<0.05).Western blot results showed that compared with the other two groups,the expression of p-Akt in LY294002 group was significantly decreased(P<0.05),and the Bax was significantly increased(P<0.05).Conclusion LY294002 effectively inhibits RKO cells and induces proliferation apoptosis by suppressing the PI3K/Akt pathway.
