Research of mechanism of betulinic acid on Va24NKT cells killing pancreatic cancer cells
Objective To study the mechanism of betulinic acid on the killing of pancreatic cancer cells by Va24NKT cells.Methods Va24NKT cells were expanded from peripheral blood,and different concentrations of betulinic acid were applied to the Va24NKT cells and SW-1990 cells.MTT method was used to detect the growth of the Va24NKT cells and SW-1990 cells,flow cytometry was used to detect the expression of NKG2D,and western blot was used to detect P-ERK1/2,ERK1/2 in the NKT cells affected by different concentrations of betulinic acid.Results The concentration of betulinic acid in 0.125~2μg/mL could promote the growth of Va24NKT cells(P<0.05).The expression of NKG2D in Va24NKT cells induced by betulinic acid was significantly higher than that in the control group(P<0.05),and the killing activity against SW-1990 cells was also significantly higher than that in the control group(P<0.05).Conclusion Betulinic acid can enhance the killing effect of Va24NKT cells on pancreatic cancer cells at a certain concentration,and the promotion of betulinic acid on the growth of Va24NKT cells is related to ERK1/2 pathway.
