响应面法优化千金黄连丸中小檗碱和梓醇的超声提取工艺

摘要
目的 优化千金黄连丸中小檗碱和梓醇的超声提取工艺.方法 采用单因素实验,考察超声时间、超声温度、超声功率、料液比对千金黄连丸中小檗碱和梓醇含量的影响.在单因素实验的基础上,以通过熵权法计算的小檗碱含量、梓醇含量的综合评分为评价指标,采用响应面法优化千金黄连丸中小檗碱和梓醇的超声提取工艺.结果 最佳工艺参数为料液比 1:30(g:mL),超声时间 50min,超声温度 27℃,超声功率 67%(167.5W,最大功率 250W),此条件下,小檗碱含量为 4.15,梓醇含量为 0.593,综合评分为 2.34.结论 响应面法操作方便、省时、重复性好,结果可靠,为千金黄连丸的进一步开发利用提供了证据.
Abstract
Objective To optimize the ultrasonic extraction process of berberine and catalpa alcohol in Qianjin Huanglian pills.Methods The effects of ultrasonic time,ultrasonic temperature,ultrasonic power and solid-liquid ratio on the contents of berberine and catalpa alcohol in Qianjin Huanglian pills were investigated by single factor experiments,and the ultrasonic extraction process of berberine and catalpa alcohol in Qianjin Huanglian pills was optimized by response surface method on the basis of single factor experiments,and the comprehensive scores of berberine and catalpa alcohol content calculated by entropy weight method were used as evaluation indexes,and the response surface method was used to optimize the ultrasonic extraction process of berberine and catalpa alcohol in Qianjin Huanglian pills.Results The optimal process parameters were as follows:solid-liquid ratio of 1:30(g:mL),ultrasonic time of 50min,ultrasonic temperature of 27℃,ultrasonic power of 67%(167.5W,maximum power 250W).Under these conditions,the berberine content was 4.15,the catalpa alcohol content was 0.593,and the comprehensive score was 2.34.Conclusion The response surface method is convenient,time-saving,reproducible and reliable,which provides evidence for the further development and utilization of Qianjin Huanglian pills.
