为克服不同发育阶段和不同形态特征的检疫性杂草少花蒺藜草的形态鉴定限制,通过设计少花蒺藜草DNA条形码matK、ITS、rbcL、psbA和trnL序列引物并对其PCR反应体系优化,比较各序列的扩增和测序效率.结果表明:matK、ITS、rbcL、psbA和trnL序列引物对少花蒺藜草的扩增和测序成功率均较高,PCR反应的最佳退火温度分别为matK 56℃、ITS 60℃、rbcL 60℃、psbA 60℃、trnL 60℃.在上述反应体系和条件下获得清晰明亮的目的条带,matK、ITS、rbcL、psbA和trnL可作为鉴别检疫性杂草少花蒺藜草DNA条形码组合的参考序列.
DNA Barcode Identification System Establishment and Optimization of Quarantine Weed Cenchrus Spinifex Cav
To overcome the limitations of morphological identification of the quarantine weed Cenchrus spinifex at different de-velopmental stages and with different morphological characteristics,the amplification and sequencing efficiencies of the sequences were compared by designing primers for the DNA barcodes matK,ITS,rbcL,psbA,and trnL sequences of Cenchrus spinifex and optimizing their PCR reaction systems.The results showed that the success rates of amplification and sequencing of Cenchrus spinifex were high with primers of matK,ITS,rbcL,psbA and trnL,and the optimal annealing temperatures for PCR reaction were matK 56℃,ITS 60℃,rbcL 60℃,psbA 60℃,trnL 60℃.The objective bands obtained under the above reaction system and conditions are clear and bright,and matK,ITS,rbcL,psbA and trnL can be used as a combination DNA barcode reference sequence to identify Cenchrus Spinifex Cav.
